=== High-resolution mass spectrometry (MS) and high-performance liquid chromatography (HPLC) === MS and HPLC are commonly applied to glycan cleaved either enzymatically or chemically from the target. In case of glycolipids, they can be analyzed directly without separation of the lipid component. N-glycans from glycoproteins are analyzed routinely by high-performance-liquid-chromatography (reversed phase, normal phase and ion exchange HPLC) after tagging the reducing end of the sugars with a fluorescent compound (reductive labeling). A large variety of different labels were introduced in the recent years, where 2-aminobenzamide (AB), anthranilic acid (AA), 2-aminopyridin (PA), 2-aminoacridone (AMAC) and 3-(acetylamino)-6-aminoacridine (AA-Ac) are just a few of them. Different labels have to be used for different ESI modes and MS systems used. O-glycans are usually analysed without any tags. Fractionated glycans from high-performance liquid chromatography (HPLC) instruments can be further analyzed by MALDI-TOF-MS(MS) to get further information about structure and purity. === Early life === McKinty was born in Belfast, Northern Ireland in 1968. The fourth of five children, he grew up in the Victoria area of Carrickfergus, County Antrim. His father was a welder and boilermaker at the Harland and Wolff shipyard before becoming a merchant seaman. He grew up reading science fiction and crime novels by the likes of Ursula Le Guin, J G Ballard and Jim Thompson. He studied law at the University of Warwick and politics and philosophy at the University of Oxford. After graduating from Oxford in 1993, McKinty moved to New York and found work in a number of occupations: security guard, barman, bookstore clerk, rugby coach, door to door salesman and librarian for the Columbia University Library. In 1999, while his wife studied for a Fulbright in Israel, McKinty played loose head prop forward for the Jerusalem Lions Rugby Club. === Haddie Braverman === Haddie Braverman (Sarah Ramos) is Adam and Kristina's oldest child. She is 15 in the beginning of the series. She is a straight-A student who plays soccer and has several friends; she aspires to be the quintessential "good girl" to compensate for her brother's behavioral problems. As the show progresses, however, she begins to assert her independence and experiment with boys and drugs. In Season 2, she volunteers in a soup kitchen where she meets Alex. They become romantically involved, but her parents object to their relationship because they feel Alex is too mature for her, with too many "grown-up" problems (he is a recovering alcoholic who left an abusive home and lives alone). Her parents' opposition to their relationship, as well as removing her right to privacy, causes Haddie to move out to live with her grandparents. The conflict is eventually resolved when Haddie returns home, and Adam and Kristina grant her permission to continue seeing her boyfriend. She loses her virginity to Alex the night of her junior prom. On another occasion, her parents find out she is having sex when she misdials them during sex. During the beginning of the third season, Alex gets arrested and charged with assault after punching another man at a party. Haddie feels personally responsible since he was only there to pick her up. Alex soon ends the relationship with her even after the charges are dropped because he feels that they are too different. She dedicates the rest of her senior year to school, and gets accepted into Cornell.
== Indications and uses == Glycated hemoglobin testing is recommended for both checking the blood sugar control in people who might be prediabetic and monitoring blood sugar control in patients with more elevated levels, termed diabetes mellitus. For a single blood sample, it provides far more revealing information on glycemic behavior than a fasting blood sugar value. However, fasting blood sugar tests are crucial in making treatment decisions. The American Diabetes Association guidelines are similar to others in advising that the glycated hemoglobin test be performed at least twice a year in patients with diabetes who are meeting treatment goals (and who have stable glycemic control) and quarterly in patients with diabetes whose therapy has changed or who are not meeting glycemic goals. Glycated hemoglobin measurement is not appropriate where a change in diet or treatment has been made within six weeks. Likewise, the test assumes a normal red blood cell aging process and mix of hemoglobin subtypes (predominantly HbA in normal adults). Hence, people with recent blood loss, hemolytic anemia, or genetic differences in the hemoglobin molecule (hemoglobinopathy) such as sickle-cell disease and other conditions, as well as those who have donated blood recently, are not suitable for this test. Due to glycated hemoglobin's variability, additional measures should be checked in patients at or near recommended goals.
