=== Class I Amidotransferase Domain === The amidotransferase domain is responsible for removal of the amide nitrogen from the glutamine substrate. The class I amidotransferase domain is made of the N terminal 206 residues of the enzyme, and consists of 12 beta strands and 5 alpha helices; the core of this domain is an open 7-stranded mixed beta sheet. Its catalytic triad includes Cys86, His181 and Glu183. His181 is a base and Glu183 is a Hydrogen bond acceptor from the Histidine imidazole ring. Cys86 is the catalytic residue and is conserved. It falls into a nucleophile elbow, where it is at the end of a beta strand and the beginning of an alpha helix, and has little flexibility in its phi and psi angles; thus, Gly84 and Gly88 are conserved and allow for the tight packing of amino acids surrounding the catalytic residue.
==== Bas-relief ==== In 1978 Joe Nickell noted that the Shroud image had a three-dimensional quality and thought its creation may have involved a sculpture of some type. He advanced the hypothesis that a medieval rubbing technique was used to produce the image, and set out to demonstrate this. He noted that while wrapping a cloth around a sculpture with normal contours would result in a distorted image, Nickell believed that wrapping a cloth over a bas-relief might result in an image like the one seen on the shroud, as it would eliminate wraparound distortions. For his demonstration, Nickell wrapped a wet cloth around a bas-relief sculpture and allowed it to dry. He then applied powdered pigment rather than wet paint (to prevent it soaking into the threads). The pigment was applied with a dauber, similar to making a rubbing from a gravestone. The result was an image with dark regions and light regions convincingly arranged. In a photo essay in Popular Photography magazine, Nickell demonstrated this technique step-by-step. Other researchers later replicated this process. In 2005 the researcher Jacques di Costanzo constructed a bas-relief of a Jesus-like face and draped wet linen over it. == Pathophysiology == Hemoglobin H disease is a genetic disorder resulting in absent or impaired production of the α-globin protein, a normal component of the hemoglobin. The disease occurs when the α-globin gene expression is reduced to less than 30% of the normal expression. In a healthy individual there are four copies of the gene which encode the α-globin protein. α-globin is encoded by the HBA1 (2 copies) and HBA2 (2 copies) genes. The genotype of healthy individuals with four normal copies of α-globin is annotated as αα/αα. In individuals with deletional HbH disease, there is deletion of three of the four α-globin alleles, which is annotated as --/-α. === Total contact casting === Total contact casting (TCC) is a specially designed cast designed to take the weight of the foot (off-loading) in patients with DFUs. Reducing pressure on the wound by taking the weight of the foot has proven to be very effective in DFU treatment. DFUs are a major factor leading to lower leg amputations among the diabetic population in the US, with 85% of amputations in diabetics being preceded by a DFU. Furthermore, the 5 year post-amputation mortality rate among diabetics is estimated at 45% for those with neuropathic DFUs. TCC has been used for off-loading DFUs in the US since the mid-1960s and is regarded by many practitioners as the "reference standard" for off-loading the bottom surface (sole) of the foot. TCC helps patients maintain their quality of life. By encasing the patient's complete foot — including the toes and lower leg — in a specialist cast to redistribute weight and pressure from the foot to the lower leg during everyday movements, patients can remain mobile. TCC redistributes pressure, protects the wound, letting damaged tissue regenerate and heal. TCC also keeps the ankle from rotating during walking, which helps prevent shearing and twisting forces that can further damage the wound. Effective offloading is a key treatment modality for DFUs, particularly those where there is damage to the nerves in the feet (peripheral neuropathy). Along with infection management and vascular assessment, TCC is a vital aspect to effectively managing DFUs. TCC is the most effective and reliable method for off-loading DFUs.

Although excluding data with huge age uncertainty would enhance the overall zircon grain age accuracy, over elimination may lower overall research reliability (decrease in size of the database).
