=== Alternatives === There is strong evidence that chlorhexidine and alcohol used to clean skin prior to surgery is better than any formulation of povidone-iodine. While the 2016 World Health Organization guidelines recommended alcohol-based CHG Chlorhexidine for surgical skin preparation based on low-to-moderate quality evidence, subsequent clinical data has clarified this comparison. A large-scale 2024 non-inferiority trial involving over 3,300 patients undergoing cardiac or abdominal surgeries found that alcohol-based povidone-iodine is non-inferior to alcohol-based CHG Chlorhexidine in preventing surgical site infections (SSIs).

Freight containers are a reusable transport and storage unit for moving products and raw materials between locations or countries. There are about seventeen million intermodal containers in the world, and a large proportion of the world's long-distance freight generated by international trade is transported in shipping containers. In addition, it is estimated that several million of these containers have now been discarded due to the shipping cost of sending them back to their port of origin. Their invention made a major contribution to the globalization of commerce in the second half of the 20th century, dramatically reducing the cost of transporting goods and hence of long-distance trade. Specialized shipping containers include: high cube containers (providing an extra 1 ft (305 mm) in height to standard shipping containers), pallet wides, open tops, side loaders, double door or tunnel-tainers, and temperature controlled containers. Another specialized container, known as Transtainer, is a portable fuel and oil freight container. The hybrid bulk fuel tank is originally intended for the construction, mining, logging and farming sectors. The tank can be used to transport and store bulk fuels as well as dangerous liquids, by road, rail and sea. Sea containers are crucial for modern logistics, offering a cost-effective storage and shipping solution. These durable containers, designed for international transportation, provide secure storage for goods with robust steel construction. Beyond shipping, they find applications in on-site storage and modular living or workspaces.
== Replication == Replication of the R1 plasmid begins at the oriRI site on the plasmid. RepA is the plasma-encoded initiator protein that binds to oriRI in order to initiate replication. RepA needs a 188-bp region of DNA at minimum in order to bind. Initiation of the leading strand, primed by DnaG, occurs at a G-type priming signal. This signal is located 400 bp downstream of the RepA-binding sequences. A newly synthesized RepA protein is used by an oriR on the same template that it was synthesized on, a cis-specific action.
Early blood culture methods were labour-intensive. One of the first known procedures, published in 1869, recommended that leeches be used to collect blood from the patient. A microbiology textbook from 1911 noted that decontamination of the draw site and equipment could take over an hour, and that due to a lack of effective methods for preserving blood, the cultures would sometimes have to be prepared at the patient's bedside. In addition to subculturing the broth, some protocols specified that the blood be mixed with melted agar and the mixture poured into a petri dish. In 1915, a blood culture collection system consisting of glass vacuum tubes containing glucose broth and an anticoagulant was described. Robert James Valentine Pulvertaft published a seminal work on blood cultures in 1930, specifying—among other insights—an optimal blood-to-broth ratio of 1:5, which is still accepted today. The use of SPS as an anticoagulant and preservative was introduced in the 1930s and 40s and resolved some of the logistical issues with earlier methods. From the 1940s through the 1980s, a great deal of research was carried out on broth formulations and additives, with the goal of creating a growth medium that could accommodate all common bloodstream pathogens. === Amplifying fluorescent polymer === Amplifying fluorescent polymer (AFP) is a promising new technology and is based on synthesized polymers which bind to explosive molecules and give an amplified signal upon detection. When compounds that are not polymers are utilized for such purpose, the quenching of the fluorescence by the traces of explosives is not detectable. When amplifying fluorescent polymer in thin films absorbs a photon of light, excited state polymers (excitons) are able to migrate along the polymer backbone and between the adjacent polymer films. These sensors were originally made in order to detect trinitrotoluene. In AFP, binding of one TNT molecule results in quenching of fluorescence significantly due to the conjugated structure of the polymers. It has been reported that in practice the polymers result in 100-1000 fold increase of amplification of the quenching response. "During its excited state lifetime, the exciton propagates by a random walk through a finite volume of the polymer film." Once TNT, or any other electron-deficient (i.e., electron accepting) molecule comes in contact with the polymer, a so-called low-energy ‘trap’ forms. "If the exciton migrates to the site of the bound electron-deficient molecule before transitioning back to the ground state, the exciton will be trapped (a non-radioactive process), and no fluorescence will be observed from the excitation event.
== Mechanism of action == As a sulfonamide antibiotic, sulfanilamide functions by competitively inhibiting (that is, by acting as a substrate analogue of) enzymatic reactions involving para-aminobenzoic acid (PABA). Specifically, it competitively inhibits the enzyme dihydropteroate synthase. PABA is needed in enzymatic reactions that produce folic acid, which acts as a coenzyme in the synthesis of purines and pyrimidines. Mammals do not synthesize their own folic acid so are unaffected by PABA inhibitors, which selectively kill bacteria. However, this effect can be reversed by adding the end products of one-carbon transfer reactions, such as thymidine, purines, methionine, and serine. PABA can also reverse the effects of sulfonamides.
