== Development == A cooperative Half-Life game was first alluded to by publisher Sierra On-Line in November 2000 with the announcement of a PlayStation 2 version of Half-Life. == Plunge freezing == The method involves ultra-rapid cooling of small tissue or cell samples to the temperature of liquid nitrogen (−196 °C) or below, stopping all motion and metabolic activity and preserving the internal structure by freezing all fluid phases solid. Typically, a sample is plunged into liquid nitrogen or into liquid ethane or liquid propane in a container cooled by liquid nitrogen. The ultimate objective is to freeze the specimen so rapidly (at 104 to 106 K per second) that ice crystals are unable to form, or are prevented from growing big enough to cause damage to the specimen's ultrastructure. The formation of samples containing specimens in amorphous ice is the "holy grail" of biological cryomicroscopy. In practice, it is very difficult to achieve high enough cooling rates to produce amorphous ice in specimens more than a few micrometres in thickness. For this purpose, plunging a specimen into liquid nitrogen at its boiling point (−196 °C) does not always freeze the specimen fast enough, for several reasons. First, the liquid nitrogen boils rapidly around the specimen forming a film of insulating N2 gas that slows heat transfer to the cryogenic liquid, known as the Leidenfrost effect. == β-Lipotropin == β-Lipotropin is a 90-amino acid polypeptide that is the carboxy-terminal fragment of POMC. It was initially reported to stimulate melanocytes to produce melanin. Birdsall and Hulme demonstrated that the C-fragment of lipotropin (β-endorphin) has a high affinity for opiate receptors in the brain, and the binding was reversed by naloxone, a classical antagonist of the opiates (Bradbury et al. 1976a). Alongside this, Feldberg found that β-endorphin administered in cat ventricles was 100 times more potent than morphine as an analgesic agent (Feldberg & Smyth 1976, 1977) and the analgesia persisted for several hours. === Urinary tract infections === A UTI affects parts of the urinary system including the urethra, bladder, and kidneys. There is about a 1% risk of UTIs in boys under two years old, and most incidents occur in the first year of life. There is good but not ideal evidence that circumcision reduces the incidence of UTIs in boys under two, and there is fair evidence that the reduction in incidence is by a factor of 3 to 10 (100 circumcisions prevent one UTI). Circumcision is most likely to benefit boys who have a high risk of UTIs due to anatomical defects and may be used to treat recurrent UTIs. There is a plausible biological explanation for the reduction in UTI risk after circumcision.
