Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. Its chain combines the first seven residues of corticotropin with a C-terminal proline-glycine-proline extension, a modification intended to slow enzymatic breakdown. The free peptide has a molecular mass near 813.9 daltons. It belongs to the class of ACTH-derived fragments studied for central nervous system activity rather than for adrenal steroid stimulation. This structural relationship to a natural hormone fragment is the usual starting point for describing the compound in the literature.
Development took place during the 1980s at institutes of the Russian Academy of Sciences, where peptide fragments of ACTH were screened for cognitive and neuroprotective effects. The compound received regulatory approval in Russia as a nasal preparation, marketed for conditions such as ischemic stroke, transient ischemic attacks, and optic nerve disorders. Registration in other countries has not followed. Most published clinical reports originate from a small number of Russian research groups, and independent replication outside that setting remains limited. The regulatory status therefore differs sharply between Russia and the rest of the world.
Laboratory work points toward modulation of neurotrophic signaling, particularly expression of brain-derived neurotrophic factor and nerve growth factor in hippocampal tissue. Studies also describe effects on monoamine turnover, inflammatory mediators, and oxidative markers. These observations come mainly from animal models and cultured cells, so the causal chain in humans is not firmly established. Whether the reported molecular changes translate into measurable clinical benefit is an open question. Reviews generally present the mechanism as plausible rather than demonstrated.
Published studies examine a fairly narrow set of endpoints. Rodent experiments commonly measure maze learning, infarct volume after induced ischemia, and tissue levels of neurotrophic factors. Clinical reports from Russian centres describe attention, memory and recovery scores in patients after stroke or transient ischemic attack. Most of those human studies are small and few have been repeated by independent groups. Outcome measures differ between studies, which limits direct comparison.
Circulation time for the peptide is short because peptidases cleave it readily. The Pro-Gly-Pro tail is thought to slow breakdown compared with the bare ACTH fragment, but the gain appears modest. Absorption after intranasal dosing is limited, and only a fraction of a dose is expected to reach the central nervous system. Laboratory concentrations therefore sit well above levels achieved systemically, a gap that complicates translation from bench findings to clinical claims.

Regulatory status varies sharply by country. Semax is registered for medical use in Russia, where it appears in formularies as a nasal solution, and it also holds registration in a small number of neighbouring states. It has no approval from the United States Food and Drug Administration or the European Medicines Agency, and it is not a scheduled controlled substance in most jurisdictions. Elsewhere it circulates mainly as laboratory material, so purity documentation comes from suppliers rather than from a national pharmacopoeia.
Semax is a synthetic peptide created in the Soviet Union during the early 1980s by researchers working in Moscow. It was built from the short adrenocorticotropic hormone fragment known as ACTH(4-10), and the chain was then extended with three additional amino acids. The resulting molecule was named semax and entered clinical use in Russia in 1994. It is generally described as a nootropic and neuroprotective agent rather than as a hormone analogue.
Solid semax is typically supplied as a lyophilised powder that is hygroscopic and sensitive to moisture, light, and repeated temperature cycling. Long-term storage of the dry peptide is generally recommended at approximately -20 degrees Celsius, while shorter working periods may use refrigeration at 2 to 8 degrees Celsius. Vials should remain tightly closed and desiccated when brought to room temperature, because condensation can damage the material before it is weighed. Dividing a batch into aliquots is preferable to thawing one container repeatedly.
Dissolution is normally performed in water, phosphate-buffered saline, or normal saline, since the peptide is freely soluble in aqueous media and is rarely handled with strong organic solvents. Solution pH should be kept near neutral, because extreme acidity or alkalinity accelerates backbone hydrolysis and encourages oxidation of the methionine side chain. Once dissolved, the material is less stable than the dry powder and is commonly divided into single-use portions and frozen. Buffers containing primary amines, such as Tris, are often avoided because of possible side reactions.
Chan School of Public Health, it is estimated that around 40% of Americans are considered obese, and 18% are considered severely obese as of 2019. Severe obesity is defined as a BMI over 35 in the study. Their projections say that about half of the US population (48.9%) will be considered obese and nearly 1 in 4 (24.2%) will be considered severely obese by 2030. New data in 2024, reveals the U.S. adult obesity rate decreased by about 2% from 2020 to 2023, according to data from the U.S. National Health and Nutrition Examination Survey. Researchers attribute this decline in part to the increasing use of weight loss drugs like Wegovy and Ozempic, now used by 6% of Americans. These medications may be helping to reverse the long-standing rise in obesity rates in the country.
=== Dual-mode === In dual-mode, the mobile and stationary phases are reversed part way through the separation experiment. This requires changing the phase being pumped through the column as well as the direction of flow. Dual-mode operation is likely to elute the entire sample from the column but the order of elution is disrupted by switching the phase and direction of flow.
5-10% of the drug (active substance); 80% of fillers, disintegrants, lubricants, glidants, and binders; and 10% of compounds which ensure easy disintegration, disaggregation, and dissolution of the tablet in the stomach or the intestine. The dissolution time can be modified for a rapid effect or for sustained release. Special coatings can make the tablet resistant to the stomach acids such that it only disintegrates in the duodenum, jejunum and colon as a result of enzyme action or alkaline pH. Pills can be coated with sugar, varnish, or wax to disguise the taste. Pharmaceutical ingredients such as APIs can also be coated with a ResonantAcoustic mixer for controlled release and taste-masking.
== History == The SIRIUS software is developed by the group of Sebastian Böcker at the Friedrich Schiller University Jena, Germany and since 2019 together with Bright Giant GmbH. SIRIUS development started in 2009 as a software for identification of the molecular formula by decomposing high-resolution isotope patterns (also called MS1 data). The name is an akronym resulting from this original purpose: Sum formula Identification by Ranking Isotope patterns Using mass Spectrometry. In 2008 the group introduced the concept of fragmentation trees for identification of the molecular formula based on fragmentation mass spectrometry data, also called tandem MS or MS2 data. Back then, identification of small molecules was approached by searching in a reference spectral library. Examples of such libraries include MassBank, METLIN, or NIST/EPA/NIH EI-MS Library. However, this is limited to known molecules with available standards that have been measured and put in a reference spectral library. For unknown molecules, identification of the molecular formula is a crucial step. In 2011/2012, the group conceived fragmentation trees as a means of structural elucidation by automatically comparing these fragmentation trees. Fragmentation pattern similarities are strongly correlated with the chemical similarity of molecules. Thus, aligning the fragmentation tree of an unknown molecule to a set of known molecules helps to elucidate its structure. Fragmentation trees were introduced in SIRIUS 2.

The Colombian government initially blamed drug lord Pablo Escobar for the murder, but journalist Steven Dudley argues that many in the UP pointed at then-Interior Minister Carlos Lemos Simmonds for publicly calling out the UP as the "political wing of FARC" shortly before the murder, while others claimed it was the result of an alliance between Fidel Castaño, members of the Colombian military, and the DAS. Pecáut and Dudley argue that significant tensions had emerged between Jaramillo, FARC, and the Communist Party due to the candidate's recent criticism of the armed struggle and their debates over the rebels' use of kidnapping, almost leading to a formal break. Jaramillo's death led to a large exodus of UP militants; in addition, by then many FARC cadres who had joined the party had already returned to clandestinity, using the UP experience as an argument in favor of revolutionary war. The M-19 and several smaller guerrilla groups were successfully incorporated into a peace process as the 1980s ended and the 1990s began, which culminated in the elections for a Constituent Assembly of Colombia that would write a new constitution, which took effect in 1991. Contacts with the FARC, which had irregularly continued despite the end of the ceasefire and the official 1987 break from negotiations, were temporarily cut off in 1990 under the presidency of César Gaviria Trujillo (1990–1994).
=== Thymus === The thymus gland is one of the first organs to degenerate in normal healthy individuals. Researchers from the University of Edinburgh have succeeded in regenerating a living organ that closely resembles a juvenile thymus in terms of structure and gene expression profile.
Bacteria can also use a NADP-dependent glyceraldehyde 3-phosphate dehydrogenase for the same purpose. Like the pentose phosphate pathway, these pathways are related to parts of glycolysis. Another carbon metabolism-related pathway involved in the generation of NADPH is the mitochondrial folate cycle, which uses principally serine as a source of one-carbon units to sustain nucleotide synthesis and redox homeostasis in mitochondria. Mitochondrial folate cycle has been recently suggested as the principal contributor to NADPH generation in mitochondria of cancer cells. NADPH can also be generated through pathways unrelated to carbon metabolism. The ferredoxin reductase is such an example. Nicotinamide nucleotide transhydrogenase transfers the hydrogen between NAD(P)H and NAD(P)+, and is found in eukaryotic mitochondria and many bacteria. There are versions that depend on a proton gradient to work and ones that do not. Some anaerobic organisms use NADP+-linked hydrogenase, ripping a hydride from hydrogen gas to produce a proton and NADPH. Like NADH, NADPH is fluorescent. NADPH in aqueous solution excited at the nicotinamide absorbance of ~335 nm (near UV) has a fluorescence emission which peaks at 445-460 nm (violet to blue). NADP+ has no appreciable fluorescence.
This is academically illustrated through the classical Jain narrative of King Yashodhara and the dough rooster. In this ethical parable, a king compromises with a demand for ritual animal sacrifice by offering a rooster made of flour. However, Jain theology dictates that because the psychological intent to kill was present, the spiritual degradation was identical to killing a living creature. Scholars note that this narrative was historically utilized to establish that the institutionalization of meat-eating corrupts the psychological purity of a society, regardless of who physically butchers the animal.
== History == Ken and Linda Rosenthal founded the St. Louis Bread Company in 1987 after Ken visited sourdough bakery-café La Boulanger in San Francisco at the insistence of his brother. After being fascinated with the process of sourdough making, he was trained to make sourdough under instruction of the owner, Roger Brunello for a year. The first location was opened in Kirkwood, Missouri. The Rosenthals invested $150,000 and received a $150,000 Small Business Administration loan. Au Bon Pain Co., a public company, purchased the St. Louis Bread Company in 1993 for $23 million. In 1997, Au Bon Pain changed the company name to Panera, from a word that has roots in the Latin word for "breadbasket" (Classical pānārium, Vulgar pānāria) and is identical to the word for "breadbasket" in Spanish and Catalan. The original name was retained for locations in Missouri. At the same time, the St. Louis Bread Company renovated its 20 bakery-cafes in the St. Louis area. In May 1999, Au Bon Pain Co. sold the Au Bon Pain chain to the firm Bruckmann, Rosser, Sherrill & Co. for $78 million to focus on the Panera Bread chain. In 2000, Panera Bread moved its headquarters to Richmond Heights, Missouri. In 2007, Panera Bread purchased a 51% stake in Paradise Bakery & Café, a Phoenix metropolitan area-based concept with over 70 locations in 10 states, predominantly in the West and Southwest, for $21.1 million. The company purchased the balance of Paradise in June 2009.

Semax is a synthetic seven-amino-acid peptide derived from a fragment of corticotropin. It is used in Russia as a nasal preparation, while elsewhere it is studied as a research compound. It is not approved as a medicine in most countries.
Its sequence corresponds to the 4-10 region of corticotropin extended by three residues. This fragment lacks the region responsible for adrenal steroid release, so it is explored for neural rather than hormonal effects. The relationship explains the compound's classification among ACTH-derived peptides.
It holds approval in Russia for several neurological indications as a nasal spray or drop formulation. Regulatory agencies in the United States and the European Union have not approved it. Products sold elsewhere are generally labeled for research use only.
It consists mainly of animal experiments and small clinical reports, with much of the clinical material published in Russian-language journals. Large independent trials are scarce. Separating reliable effects from chance findings is consequently difficult.
