== History == Used on a short-term basis clinically to treat obesity, some appetite suppressants are also available over-the-counter. Several appetite suppressants are based on a mix of natural ingredients, mostly using green tea as its basis, in combination with other plant extracts, such as fucoxanthin, found naturally in seaweed. Drugs of this class are frequently stimulants of the phenethylamine family, related to amphetamine. The German and Finnish militaries issued amphetamines (Pervitin) to soldiers commonly during the Second World War. Similarly, the UK military was supplied with more than 72 million Benzedrine tablets and the US military with an approximately equal amount for situations, in which fatigue was not deemed to be an acceptable option. Following the war, large amphetamine surpluses were redirected for use on the black and the civilian market. Indeed, amphetamine itself was sold commercially as an appetite suppressant until it was outlawed in most parts of the world in the late 1950s because of safety issues. Many amphetamines produce side effects, including addiction, tachycardia and hypertension, making prolonged unsupervised use dangerous.
== Uses == P. rubens is the source of β-lactam antibiotics, principally of penicillins and to a lesser extent, cephalosporins. The species produces three penicillins, benzylpenicillin (G), phenoxymethylpenicillin (V) and octanoylpenicillin (K). Penicillin G is a naturally occurring compound that was first isolated and used as an antibiotic. Penicillins from P. rubens are effective against Gram-positive bacteria such as the species of Bacillus, Clostridium, Corynebacterium, Pneumococcus, Streptococcus, and Staphylococcus. They are clinically used to treat actinomycosis, anthrax, botulism, clostridial infections, diphtheria, empyema, endocarditis, food poisoning (due to Listeria), gas gangrene, meningitis, pneumonia, pericarditis, septicaemia, and syphilis. However, they are useless against Gram-negative bacteria since these bacteria are impermeable to the compounds because of their additional outer cell membrane, as well as some species producing pencillases (β-lactamases) that destroy the penicillin structure (β-lactam ring). However, semisynthetic penicillins like aminopenicillins (ampicillin, amoxicillin, and bacampicillin), carboxypenicillins (carbenicillin and ticarcillin) and ureidopenicillins (mezlocillin, azlocillin, and piperacillin) have been developed that are effective against these bacteria. Cephalosporins, naturally produced by other fungi belonging to the genus Acremonium, are one of the most widely used antibiotics by clinicians and is also produced by P. rubens. These studies found that, in free enzyme, the zinc coordination number is five; the metal center is coordinated with two imidazole Nδ1 nitrogens, the two carboxylate oxygens of glutamate-72, and a water molecule to form a distorted tetrahedral. However, once ligand binds at the active site of carboxypeptidase A, this coordination number can vary from five to six. When bound to dipeptide glycyl-L-tyrosine, the amino nitrogen of the dipeptide and the carbonyl oxygen replaced the water ligand. This would yield a coordination number of six for the zinc in the carboxypeptidase A- dipeptide glycyl-L-tyrosine complex. Electron density maps gave evidence that the amino nitrogen occupies a second position near glutamate-270. The closeness of these two residues would result in a steric hindrance preventing the water ligand from coordinating with zinc. This would result in a coordination number of five. Data for both are substantial, indicating that both situations occur naturally. There are two proposed mechanisms for the catalytic function of carboxypeptidase A. The first is a nucleophilic pathway involving a covalent acyl enzyme intermediate containing active site base Glu-270. Evidence for this anhydride intermediate is mixed; Suh and colleagues isolated what is assumed to by the acyl intermediate.
