The short version of Lyophilized powder fits in a sentence. The long version — which is the one that helps — is below.
This page was last updated on 2025-10-04 and is reviewed periodically as new material appears.
Stability depends heavily on physical state. Lyophilized powder held dry, cold and dark retains its content over long periods, whereas dissolved peptide begins to change within days at room temperature. The most cited degradation route is oxidation of the methionine residue, which converts the peptide to a sulfoxide form that elutes differently on chromatography. Hydrolysis of amide bonds and adsorption onto container walls contribute smaller losses. Buffers that exclude oxygen from the headspace slow the oxidation pathway, but no single condition prevents all change indefinitely.
Practical handling follows from those properties. Bulk material is best divided into single-use portions soon after receipt, because each thaw exposes the whole container to moisture and temperature cycling. Vials should be allowed to reach room temperature before opening to prevent condensation on the powder. Low-binding plasticware reduces loss of dilute solutions, and sterile filtration is used when a preparation must remain free of microbial growth. Records of batch number, reconstitution date and storage history are what allow a later analytical result to be interpreted meaningfully.
Identity and purity of Semax are established mainly by reversed-phase high-performance liquid chromatography coupled with mass spectrometry. The chromatographic trace gives a purity estimate as a percentage of total peak area, while electrospray or matrix-assisted laser desorption ionization confirms the molecular mass against the calculated value. Amino acid analysis and sequence-specific fragmentation provide further confirmation when a supplier's chain of custody is unclear. Vendors frequently quote a purity figure without stating the detection wavelength or the integration method, which limits how far one number can be compared with another.
Semax is a synthetic seven-amino-acid peptide whose sequence extends the ACTH(4-10) fragment with a C-terminal proline-glycine-proline tripeptide. The commonly cited sequence is Met-Glu-His-Phe-Pro-Gly-Pro, giving a molecular formula near C37H51N9O10S and a molecular weight close to 813.9 g/mol. It belongs to the broader class of synthetic ACTH fragments studied for central nervous system effects rather than for adrenal steroid stimulation. In practice the material appears as a lyophilized white powder for laboratory work or as a dilute saline solution in clinical settings.
Development is attributed to researchers at the Institute of Molecular Genetics in Moscow during the early 1980s, building on earlier Soviet work with ACTH fragments. Russian regulatory approval followed for intranasal use, and the compound has remained commercially available there for decades. Most published human data originate from Russian and, later, some Eastern European clinical reports, which are not always accessible in English translation. Outside that region the material is generally handled as a research chemical rather than a licensed medicine.
Regulatory status differs sharply between jurisdictions. In Russia the peptide is registered as a prescription nasal preparation, while agencies such as the United States Food and Drug Administration have not approved it for any indication. Products sold elsewhere are typically labeled for laboratory research only, and such labels shift responsibility for safe handling to the purchaser. Because the same name covers pharmaceutical-grade nasal drops and bulk research powder, identity and purity documentation becomes the main practical concern when comparing sources.

Proposed mechanisms center on neurotrophic signaling rather than on classical melanocortin receptor activation. Rodent experiments have reported shifts in the expression of brain-derived neurotrophic factor and nerve growth factor after administration, together with changes in the associated receptor systems. Several authors argue that the peptide acts largely through its degradation products and their interaction with peptidergic pathways, but this remains a hypothesis rather than a settled finding. No single molecular target has been identified in a way that the field broadly accepts.
Published research covers ischemic stroke, traumatic brain injury, cognitive impairment, optic nerve conditions and attention-related measures. Much of the human evidence comes from small trials conducted in one country, which limits how far the results generalize. Animal models supply the larger share of the data, and effects seen in rodents do not transfer automatically to people. Reviews have noted that methodological reporting is often incomplete, making it difficult to pool results or compare treatment schedules across studies.
Pharmacokinetic accounts emphasize rapid breakdown. After intravenous dosing the intact peptide disappears from blood within minutes, and nasal delivery produces low but measurable concentrations. Metabolites rather than the parent molecule may account for part of the observed activity, although the relative contribution is unresolved. Dosing in the literature varies widely and no optimal schedule has been agreed. These gaps are regularly cited as a reason the findings have not produced broad clinical adoption beyond the original research setting.
=== Cleavage products === LL-37 is cleaved into a number of smaller fragments which retain anti-microbial and anti-cancer effects but generally have a lower toxicity to human cells. RK-31, KS-30 and KR-20 are naturally occurring fragments, while other related peptides have been made synthetically based on natural fragments of LL-37 during research into cathelicidins, and in some cases have amino acid substitutions.
inborn error of metabolism Any genetically determined biochemical variation affecting one or more metabolic functions in a way that causes a congenital disease or disorder in humans. Most such errors are due to mutations in single genes encoding enzymes that facilitate important metabolic reactions such as the conversion of specific substrates into unique products, which may affect numerous downstream steps in one or more metabolic pathways and thereby contribute to a diverse variety of symptoms; often they cause the accumulation of toxic intermediates or impair the body's ability to synthesize essential compounds.
=== Nixon's key decision maker === On 17 February 1969, Nixon then told the Soviet ambassador Anatoly Dobrynin that all matters of substance were to go through Kissinger rather than the Secretary of State William Rogers. Shortly afterwards, Kissinger met with Dobrynin to tell him that Nixon would not accept any settlement that looked like a defeat nor did he want any change in the regime in Saigon, though "evolution" of the Saigon regime was acceptable. Dobrynin, who served in Washington for many years, had a favorable impression of Kissinger, who was not dogmatic and rigid like his predecessor W.W. Rostow nor dull and unimaginative like Dean Rusk. Kissinger then set about undermining Henry Cabot Lodge Jr., the head of the American peace delegation in Paris, as he asked Dobrynin to set up a secret meeting in Paris between him and Le Duc Tho, the most important member of the North Vietnamese delegation in Paris. On 22 February 1969, the Viet Cong launched an offensive in South Vietnam, which Kissinger called "an act of extraordinary cynicism". Nixon, on a trip to Europe, took the offensive as a personal insult and wanted to bomb Cambodia in retaliation. Kissinger persuaded Nixon to wait until his European trip was over. As part of the "linkage" concept, Kissinger in March 1969 sent Cyrus Vance to Moscow with the message that if the Soviet Union pressured North Vietnam into a diplomatic settlement favorable to the United States, the reward would be concessions on the talks on limiting the nuclear arms race.

In addition to CYP2B6, CYP2A6 may be involved in the metabolism of selegiline to a lesser extent. Birth control pills containing the synthetic estrogen ethinylestradiol and a progestin like gestodene or levonorgestrel have been found to increase peak levels and overall exposure to oral selegiline by 10- to 20-fold. High levels of selegiline can lead to loss of MAO-B selectivity and inhibition of MAO-A as well. This increases susceptibility to side effects and interactions of non-selective monoamine oxidase inhibitors (MAOIs), such as tyramine-induced hypertensive crisis and serotonin toxicity when combined with serotonergic medications. However, this study had a small sample size of four individuals as well as other methodological limitations. The precise mechanism underlying the interaction is unknown, but is likely related to cytochrome P450 inhibition and consequent inhibition of selegiline first-pass metabolism by ethinylestradiol. In contrast to birth control pills containing ethinylestradiol, menopausal hormone therapy with estradiol and levonorgestrel did not modify peak levels of selegiline and only modestly increased overall exposure (+59%). Hence, menopausal hormone therapy does not pose the same risk of interaction as ethinylestradiol-containing birth control pills when taken together with selegiline. Overall exposure to selegiline with oral selegiline has been found to be 23-fold lower in people taking anticonvulsants known to strongly activate drug-metabolizing enzymes. The anticonvulsants included phenobarbital, phenytoin, carbamazepine, and amobarbital.
=== April === 1 April An entirely new class of antibiotics with potent activity against multi-drug resistant bacteria is discovered. These compounds target a protein called LpxH, and are shown to cure bloodstream infections in mice. An analysis of the European Union's Common Agricultural Policy suggests 82% of the EU's agricultural subsidies are used for animal-based foods which "are associated with 84% of embodied greenhouse gas emissions of EU food production".
=== Diagnosis === It is difficult to differentiate the effects of low level metal poisoning from the environment with other kinds of environmental harms, including nonmetal pollution. Generally, increased exposure to heavy metals in the environment increases the risks for several diseases.
==== Diagnostic Statistical Manual assertion ==== DSM-IV lists General diagnostic criteria for a personality disorder guideline that "The pattern must not be better accounted for as a manifestation of another mental disorder, or to the direct physiological effects of a substance (e.g. drug or medication) or a general medical condition (e.g. head trauma).". As a result, AAS users may get misdiagnosed by a psychiatrist not told about their habit.

== Management == Opioid use disorders typically require long-term treatment and care with the goal of reducing the person's risks and improving their long-term physical and psychological condition. First-line management involves the use of opioid replacement therapies, particularly methadone, naltrexone, morphine, hydromorphone, buprenorphine/naloxone (Suboxone), and diamorphine (heroin), which is the most effective treatment option of all drugs. Withdrawal management alone is strongly discouraged, because of its association with elevated risks of HIV and hepatitis C transmission, high rates of overdose deaths, and nearly universal relapse. This approach is seen as ineffective without plans for transition to long-term evidence-based addiction treatment, such as opioid agonist treatment. Though treatment reduces mortality rates, the first four weeks after treatment begins and the four weeks after treatment ceases are the riskiest times for drug-related deaths. These periods of increased vulnerability are significant because many of those in treatment leave programs during these periods. There is evidence that people with opioid use disorder who are dependent on pharmaceutical opioids may require a different management approach from those who take heroin.
RTI-5152-12, or WW-12 (in patent), is a synthetic small-molecule agonist of the atypical chemokine receptor ACKR3 (CXCR7) that was derived from the naturally occurring alkaloid conolidine. RTI-5152-12 has 15-fold improved potency towards ACKR3 relative to conolidine. ACKR3 is a novel opioid receptor which functions as a broad-spectrum trap or scavenger for endogenous opioid peptides, including enkephalins, dynorphins, and nociceptin. The receptor acts as a negative modulator of the opioid system by decreasing the availability of opioid peptides for their classical receptors like the μ-opioid receptor. Ligands of ACKR3, by competitively displacing endogenous opioid peptides from ACKR3, can potentiate the actions of these endogenous opioids and produce effects like analgesia and anxiolysis in animals. RTI-5152-12 is being developed as a potential pharmaceutical drug and, as of December 2021, is in the preclinical stage of development for treatment of pain. The chemical structure was not disclosed until a patent was published in June 2022.
Novel ingredients must undergo scientific assessment before being considered safe for consumption, and a safety assessment report must be completed for ingredient authorization. In addition, all pet food animal products must be considered fit for human food consumption, a measure put into place to prevent the spread of zoonotic diseases, and plants that will be manufacturing the pet food must be authorized under two of the EU legislature bodies. To learn more about labelling and regulations of pet food, see Pet food.

Suppliers normally quote a percentage of total chromatographic peak area, most often from reversed-phase HPLC. That figure says nothing about what the remaining percentage contains, and it depends on the detection wavelength used. A mass spectrometry result is a separate and stronger check on identity.
The methionine residue at the start of the chain is vulnerable to oxidation, producing a sulfoxide variant. Amide bonds can also hydrolyze, though more slowly under neutral conditions. Cold storage, oxygen exclusion and minimization of freeze-thaw cycles slow both processes but do not stop them.
A purity number on a certificate does not establish that a powder is the same product as a registered nasal medicine. Counter-ion content, residual solvents and peptide-related impurities may differ between the two. Independent verification is the only way to narrow that gap.
It is a short synthetic peptide built from seven amino acids: methionine, glutamic acid, histidine, phenylalanine and three prolines. The sequence derives from the 4-10 fragment of adrenocorticotropic hormone with an added proline-glycine-proline tail. No plant or animal extract is involved; the material is produced by solid-phase peptide synthesis.
