intranasal delivery raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.
This page was last updated on 2026-07-15 and is reviewed periodically as new material appears.
The mechanisms attributed to semax are inferred from animal and cell studies rather than traced to one confirmed target. The most frequently cited pathway involves increased expression of brain-derived neurotrophic factor and nerve growth factor in hippocampal and cortical tissue. Some work points to engagement of melanocortin receptors, particularly MC4, which the parent ACTH fragment can activate. Effects on dopaminergic and serotonergic signalling have also been reported. No single account explains all observed results, and the relative weight of each pathway remains unsettled.
Published studies examine a fairly narrow set of endpoints. Rodent experiments commonly measure maze learning, infarct volume after induced ischemia, and tissue levels of neurotrophic factors. Clinical reports from Russian centres describe attention, memory and recovery scores in patients after stroke or transient ischemic attack. Most of those human studies are small and few have been repeated by independent groups. Outcome measures differ between studies, which limits direct comparison.
Circulation time for the peptide is short because peptidases cleave it readily. The Pro-Gly-Pro tail is thought to slow breakdown compared with the bare ACTH fragment, but the gain appears modest. Absorption after intranasal dosing is limited, and only a fraction of a dose is expected to reach the central nervous system. Laboratory concentrations therefore sit well above levels achieved systemically, a gap that complicates translation from bench findings to clinical claims.

Published research covers ischemic stroke, traumatic brain injury, cognitive impairment, optic nerve conditions and attention-related measures. Much of the human evidence comes from small trials conducted in one country, which limits how far the results generalize. Animal models supply the larger share of the data, and effects seen in rodents do not transfer automatically to people. Reviews have noted that methodological reporting is often incomplete, making it difficult to pool results or compare treatment schedules across studies.
Pharmacokinetic accounts emphasize rapid breakdown. After intravenous dosing the intact peptide disappears from blood within minutes, and nasal delivery produces low but measurable concentrations. Metabolites rather than the parent molecule may account for part of the observed activity, although the relative contribution is unresolved. Dosing in the literature varies widely and no optimal schedule has been agreed. These gaps are regularly cited as a reason the findings have not produced broad clinical adoption beyond the original research setting.
Identity and purity of Semax are established mainly by reversed-phase high-performance liquid chromatography coupled with mass spectrometry. The chromatographic trace gives a purity estimate as a percentage of total peak area, while electrospray or matrix-assisted laser desorption ionization confirms the molecular mass against the calculated value. Amino acid analysis and sequence-specific fragmentation provide further confirmation when a supplier's chain of custody is unclear. Vendors frequently quote a purity figure without stating the detection wavelength or the integration method, which limits how far one number can be compared with another.
Stability depends heavily on physical state. Lyophilized powder held dry, cold and dark retains its content over long periods, whereas dissolved peptide begins to change within days at room temperature. The most cited degradation route is oxidation of the methionine residue, which converts the peptide to a sulfoxide form that elutes differently on chromatography. Hydrolysis of amide bonds and adsorption onto container walls contribute smaller losses. Buffers that exclude oxygen from the headspace slow the oxidation pathway, but no single condition prevents all change indefinitely.
Practical handling follows from those properties. Bulk material is best divided into single-use portions soon after receipt, because each thaw exposes the whole container to moisture and temperature cycling. Vials should be allowed to reach room temperature before opening to prevent condensation on the powder. Low-binding plasticware reduces loss of dilute solutions, and sterile filtration is used when a preparation must remain free of microbial growth. Records of batch number, reconstitution date and storage history are what allow a later analytical result to be interpreted meaningfully.
Biological carbon fixation, or carbon assimilation, is the process by which living organisms convert inorganic carbon (particularly carbon dioxide, CO2) to organic compounds. These organic compounds are then used to store energy and as structures for other biomolecules. Carbon is primarily fixed through photosynthesis, but some organisms use chemosynthesis in the absence of sunlight. Chemosynthesis is carbon fixation driven by chemical energy rather than from sunlight. The process of biological carbon fixation plays a crucial role in the global carbon cycle, as it serves as the primary mechanism for removing CO2 from the atmosphere and incorporating it into living biomass. The primary production of organic compounds allows carbon to enter the biosphere. Carbon is considered essential for life as a base element for building organic compounds. The flow of carbon from the Earth's atmosphere, oceans and lithosphere into lifeforms and then back into the air, water and soil is one of the key biogeochemical cycles (or nutrient cycles). Understanding biological carbon fixation is essential for comprehending ecosystem dynamics, climate regulation, and the sustainability of life on Earth. Organisms that grow by fixing carbon, such as most plants and algae, are called autotrophs. These include photoautotrophs (which use sunlight) and lithoautotrophs (which use inorganic oxidation). Heterotrophs, such as animals and fungi, are not capable of carbon fixation but are able to grow by consuming the carbon fixed by autotrophs or other heterotrophs.

=== Anti-competitive practices === AbbVie has been accused of using anti-competitive patent thickets to prevent potentially cheaper biosimilars from entering the market. AbbVie and Alvotech, filed lawsuits against each other regarding a Humira biosimilar introduced by Alvotech. The lawsuits were settled out of court in 2022. Forest Laboratories, a subsidiary of AbbVie, was accused of using unlawful deals to prevent generic versions of its Alzheimer's disease drug, Namenda, from entering the market. In 2018, AbbVie agreed to pay $25 million to resolve allegations that it used kickback schemes to promote its cholesterol drug TriCor (fenofibrate). In 2020, AbbVie agreed to pay $24 million to resolve allegations that it used kickback schemes to promote Humira using "nurse ambassadors". A report from the House Oversight and Reform Committee found that "AbbVie pursued a variety of tactics to increase drug sales while raising prices for Americans, including exploiting the patent system to extend its market monopoly, abusing orphan drug protections to further block competition, and engaging in anticompetitive pricing practices". Pursuant to the Inflation Reduction Act, the price of Imbruvica was reduced by 38%, to $9,319 per month, effective in 2026.

Ernest William Beckett (1856–1917), who became the 2nd Baron Grimthorpe. Helen Louisa Beckett-Denison (1858–1935). Adeline Gertrude Beckett-Denison (1859–1902), who married Sir Frederick Milner, 7th Baronet. Violet Katharine Beckett-Denison (1860–1883), who married Reginald Walkeline Chandos-Pole, grandson of Leicester Stanhope, 5th Earl of Harrington. Maud Augusta Beckett-Denison (1864–1927), who married Lord Henry Nevill (later 3rd Marquess of Abergavenny). William Gervase Beckett (1866–1937), who became Sir Gervase Beckett, 1st Baronet of Kirkdale Manor. Hon. Rupert Evelyn Beckett (1870–1955), who married Muriel Helen Florence Paget, granddaughter of Henry Paget, 2nd Marquess of Anglesey. Beckett died at the age of 64 when he fell under a train at Wimborne. He had arrived at Wimborne railway station to change trains for Bournemouth, and while waiting, he went for a walk. He was walking beside the track on the way back to the station when a strong wind blew his hat off, and he fell almost immediately under the train. His body was cut to pieces and carried some 56 feet. Following his death, Mrs. Gertrude Beatrice Brinkworth alleged she was his mistress and mother of his adopted children and sued his executors. Some money was paid, but the case came before the court, and the jury declared Mrs. Brinkworth's case to be a 'swindle' and rejected her claims even before the evidence in the case had been completed.
=== Possible future treatments === A peptide that prevents and treats citrus greening disease in greenhouse trials was being tested in field trials in 2021; an enhanced injectable version of the product was being developed in 2020. Two types of antisense oligonucleotide (FANA and Morpholinos) can be delivered efficiently into citrus trees, suppressing their RNA targets. FANA can suppress 'Candidatus Liberibacter asiaticus' in citrus trees. Morpholinos can suppress CLas in infected citrus trees and the psyllid vectors. Furthermore, the PPMOs designed to endosymbiotic bacteria of the psyllid vectors, can reduce psyllid populations by targeting and suppressing the insects endosymbionts, the bacteria which are essential for psyllid survival. Morpholinos must be covalently linked with a charged molecule or peptide, to enter bacteria. The target RNA is made susceptible to cleavage by ribonuclease P (RNase-P). Researchers at the Chinese Academy of Sciences found that the E3 ubiquitin ligase PUB21 acts as an essential susceptibility gene, marking the MYC2 transcription factor for degradation. Introduction of a dominant negative variant of PUB21 was successful in conferring resistance to HLB by stabilizing MYC2 levels.
== Role as a cancer promoter == FGF19 is frequently amplified in human cancers. Amplification of the FGF19 genomic locus was found in liver cancer, breast cancer, lung cancer, prostate cancer, bladder cancer, and esophageal cancer, among others. Targeting FGF19 inhibits tumor growth in colon cancer cells and hepatocellar carcinoma. Increase in FGF19 correlates with tumor progression and poorer prognosis of hepatocellular carcinoma.
== Research directions == The approach of screening the soil for useful compounds using genomics has been done by others, and is likely to continue to be pursued as a method to further explore primary metabolites and secondary metabolites made by microorganisms. As of February 2018, the malacidins had not been tested on humans. At the time of their discovery it was unknown whether the discovery would lead to any new antibiotic drugs; showing that a potential drug is safe and effective takes years of work and millions of dollars, and the scientists said at the time that they had no plans to try to develop a drug based on the work. In the 2018 paper, malacidins were shown to kill only Gram-positive bacteria and not Gram-negative bacteria. They were, however, able to kill multidrug-resistant pathogens, including bacteria resistant to vancomycin in the laboratory, and methicillin-resistant Staphylococcus aureus (MRSA) skin infections in an animal wound model. Brady, Hover, and two other authors disclosed in the 2018 paper that they had "competing financial interests, as they are employees or consultants of Lodo Therapeutics." Lodo was founded in 2016 out of Brady's laboratory, to discover new chemicals in nature as starting points for drug discovery.

It consists mainly of animal experiments and small clinical reports, with much of the clinical material published in Russian-language journals. Large independent trials are scarce. Separating reliable effects from chance findings is consequently difficult.
A fraction of an intranasal dose is thought to reach the central nervous system, and this route is favoured partly for that reason. The proportion is not well characterized. Direct measurement in humans remains difficult with current methods.
Raised BDNF and NGF expression has been reported across several animal studies, which makes the signal fairly consistent. Whether the same change occurs in humans at usable doses is not established. It is best treated as a working hypothesis rather than a confirmed clinical mechanism.
一般不建议常温长期保存。多数说明指向 -20 °C 冷冻避光。常温运输通常被视为短期可接受,但会加快降解风险。
